How digestion, indispensable amino acids and the food matrix shape protein quality—and why no single score tells the whole nutrition story.
The protein number on a nutrition information panel tells you how many grams a serving contains. It does not tell you which amino acids are present, how readily they are digested or what other nutrients arrive with the food.
Protein digestibility helps fill in part of that picture. It describes the proportion of dietary protein—or, more precisely, its amino acids—that becomes available for absorption after digestion. But digestibility is not a stand-alone verdict on whether one food or diet is ‘better’ than another.

What Is Protein Digestibility?
Protein digestibility is an estimate of how much protein or how many amino acids disappear from the digestive tract and are considered available for absorption. The definition depends on where and how it is measured.
Older approaches often use faecal nitrogen, which reflects what remains after the large intestine. Ileal digestibility measures disappearance by the end of the small intestine and better represents amino acids available to the body before colonic microbes modify the residue.
Digestibility is not the same as the body turning every absorbed amino acid into muscle. After absorption, amino acids enter metabolic pools and may be used for proteins, other nitrogen-containing compounds, oxidation or energy.

What Influences Digestibility?
Protein structure and source
Proteins differ in shape, cross-linking and accessibility to digestive enzymes. Many animal proteins test as highly digestible, while results among plant foods vary widely. These are averages for foods or test conditions, not a reason to dismiss nutrient-rich plant sources.
The food matrix
Cell walls, fibre, starch, fat and interactions between compounds can alter how enzymes reach protein. Isolated protein and the whole food from which it came may therefore produce different digestibility values.
Preparation and processing
Cooking can unfold proteins and inactivate some protease inhibitors, improving digestibility in foods such as legumes. Soaking, germination and fermentation can also alter the matrix. The Digestible Indispensable Amino Acid Score compares each digestible indispensable amino acid in a protein with a reference amino-acid pattern. The lowest ratio—the limiting amino acid—determines the score.
The Food and Agriculture Organization recommended true ileal digestibility for each indispensable amino acid, preferably measured in humans and otherwise using validated animal models. Reference patterns differ by life stage, so a score is not context-free.
DIAAS improves on the older Protein Digestibility-Corrected Amino Acid Score in important ways: it uses ileal rather than faecal digestibility, considers individual amino acids and does not automatically truncate scores above 100.
What DIAAS Cannot Tell You

DIAAS is mainly a research and food-evaluation tool. Consumers will rarely see a validated DIAAS value on a label, and scores reported online may use different datasets or test preparations.
Animal and Plant Proteins: A More Useful Comparison
Some animal-derived proteins provide all indispensable amino acids in favourable proportions and are highly digestible. Plant foods may have a lower proportion of one or more indispensable amino acids or a matrix that reduces measured digestibility, but they also contribute fibre, micronutrients and other food components.
A varied diet can combine complementary patterns—for example, legumes tend to be richer in lysine than grains, while grains may contribute more methionine relative to lysine. These foods do not need to be combined in precise ratios at every meal when the total diet is adequate.

Why Protein Grams Still Matter
A highly digestible food cannot meet protein needs if too little is eaten. Conversely, a diet with generous total protein may still be poorly balanced if it relies heavily on a source with a limiting indispensable amino acid. Quantity and quality work together.
